Humanized TNF-Alpha Mouse Model for In Vivo Preclinical Studies

Applications: inflammation and autoimmune disease

The humanized TNF-α mouse model allows in vivo efficacy and safety assessment of anti-human TNF-α compounds in the field of autoimmune diseases and inflammation.

Model features:

  • Humanization by gene substitution; the murine TNF-α gene is inactivated and replaced by the human TNF-α genomic portion
  • Human TNF-α expression is driven by the mouse TNF-α endogenous promoter (physiological TNF-α regulation and expression pattern)
  • Physiological expression level of human TNF-α (Fig. 1B)
  • No expression of murine TNF-α (Fig. 1A)
  • No physiological deregulation reported

Model validation

Mean concentrations of murine TNF-α (A) and human TNF-α (B) measured by ELISA from plasma samples collected from WT (n=2) and KI (n=2) mice treated with saline solution vs. LPS.
