Research summary

Using CRISPR/Cas9, the authors inserted an IRES‑Cre cassette downstream of the STOP codon in exon 17 of Prkcd in rats. Cre expression is selective to PKCδ⁺ cells in the central amygdala. Anatomical tracing, electrophysiology, and behavioral assays including food intake and real‑time place preference validate functionality of the model.

Key outcome of the study

Cre is expressed in PKCδ⁺ neurons; projection patterns match known circuits; optogenetic stimulation of PKCδ⁺ neurons in CeA modulates feeding latency and aversion without altering locomotion or baseline behaviors
