A Quick Knockin rat line defines a model in which a gene of interest is inserted in the permissive (neutral) locus Rosa26. The transgene can be constitutively or inducibly overexpressed.

Targeting Rosa26 enables the control of the gene copy number and gene expression, and avoids silencing or deregulation of neighboring genes.

Strengths of Quick Knockin rat models

  • Stable and predictable expression
  • Almost no position effect variegation
  • Almost no transgene silencing
  • No deregulation of neighbouring gene

Limitations of Quick Knockin rat models

  • Exogenous regulation may result in lower physiological relevancy
  • Limited physiological relevancy when multiple isoforms have been described
  • Problem of stoichiometry if interaction with other partners for signaling
    → Must be solved by an in-depth scientific analysis and a rationale risk assessment prior to model development, all included in our project feasibility assessment
  • Not functional for expression of non-dominant mutations
    → Choose alternative approaches such as, e.g., point mutation Knockin
