Baseline validation

  • hCD3 and hCD137 expression pattern in genO-panhCD3/hCD137 mice recapitulates mCD3 and mCD137 expression pattern in wild-type mice

    (A) CD3 staining on freshly isolated splenocytes. Gating on single live TCRβ+ CD4+CD8- CD25- FoxP3- (Conv CD4), TCRβ+ CD4+ CD8-CD25+ FoxP3+ (Treg) and TCRβ+ CD4- CD8+ (CD8).

    (B) Splenocytes were activated with anti-mouse CD3 and anti-mouse CD28 in presence of rm-IL-2 for 48 hours and CD137 expression was assessed. Gating on live TCRb+CD19- (TCRb cells), live CD4+CD8-CD25-FoxP3- (Conv CD4), CD4+CD8-CD25+FoxP3+ (Treg), CD4-CD8+ (CD8).

    Anti-mouse CD3: clone 145-2C11, anti-human CD3: clone OKT3.

    Anti-mouse CD137: clone 4B4-1, anti-human CD137: clone 17B5.
