Freshly isolated splenocytes from WT or genO-hTIGIT mice were activated by αCD3/αCD28 in presence of mIL-2 for 48h. Gated on live CD3+CD4+CD8-D25+FoxP3+ (Treg cells), CD3+CD4+CD8-FoxP3- (ConvCD4+), CD3+ CD4-CD8+ (CD8+ cells) and CD3-NK1.1+ (NK cells).
A) Isolated T cells from WT and genO-hTIGIT mice were labelled with CTV and activated with αCD3 in presence of rmCD155 or control. Cell proliferation was assessed by flow cytometry after 4 days. Cells gated on single live CD3+ CD4+ CD8- T cells. 2way ANOVA test, ** p < 0,005 ,*** p < 0,001 vs. αCD3
B) Isolated T cells from WT and genO-hTIGIT mice were activated with αCD3 in presence of rmCD155 or control. Secreted IFN-γ was quantified in the supernatant by ELISA after 4 days. 2way ANOVA test, p < 0,001 vs. αCD3
As reported, mCD155 cross-reacts with hTIGIT (Staniestky et al., Eur. J. Imm., 2013).
genO-hTIGIT mice were engrafted with 1x106 MC38 cells (s.c.). Once tumors reached 100 mm3, genO-hTIGIT mice were treated with anti-mPD-1 (10 mg/kg), Tiragolumab (40 mg/kg) or combotherapy, 2 times a week for 3 weeks. Tumor volume was monitored 2 to 3 times a week until tumor volume reached 1200 mm3.
