Applications of the genO‑hVISTA/hFcγR mouse model
Useful applications for researchers working in different biomedical fields, such as immuno-oncology, inflammation and autoimmunity:
- Assess the efficacy of VISTA-targeting antibodies and understand the contribution of the Fc region:
- Perform tumor growth inhibition assays in the context of monotherapies or combotherapies
- Ex vivo assessment of antibody-dependent cellular cytotoxicity (ADCC) induced by different monoclonal antibodies
- Rank Fc-engineered antibodies with enhanced or reduced binding to FcγR
- Gain insight into the mechanism of action (MOA) of diverse constructs and their potential therapeutic benefit
- Evaluate the safety and toxicity of compounds
- Analyze the PK profile and the biodistribution of hVISTA-targeting antibodies with an Fc-competent domain
- Suitable for toxicological assessments, including those relating to reproductive toxicity, as an alternative to non-human primate (NHP) models
Features
This model combines the features of two extensively validated models: genO-hVISTA and genO-hFcγR.
- genO‑hFcγR/hVISTA mice have a human-like FcγR expression pattern and physiological expression of humanized VISTA
- Expression of the following humanized receptors: FcγRI (CD64), FcγRIIA (CD32A), FcγRIIB (CD32B), FcγRIIIA (CD16A), FcγRIIIB (CD16B)
- Functional Fcγ receptors enable the assessment of Fc-mediated effector functions
- No expression of murine FcγR nor murine VISTA
- Fully immunocompetent model
